(Summary description)Alzheimer's disease is associated with intraparenchymal growth of slab amyloid deposits, amyloid plaques are the progressive deposition and transformation of soluble amyloid β-protein monomers into insoluble and fibrous aggregates, a process described as "Seed aggregation" of monomers with slow nucleation and fast growth kinetics. So far, the occurrence and development of Alzheimer's disease and the development of therapeutic methods are still difficult problems in the academic circle. In 1998, Nature Medicine published the work of M. Pitschke's team at the German Institute of Biophysics to study β-amyloid aggregation using FCS technology. This study remains a good model for follow-up research to this day. In the present study, the authors used FCS to detect the accumulation of individual amyloid-β peptides in the CSF of Alzheimer's disease patients. Soluble amyloid β-protein monomers generally exist in the extracellular space and cerebrospinal fluid, while insoluble aggregates are currently only available in brain tissue through biopsy and autopsy. In the experiment, the main amino groups of the Aβ1-40 and Aβ1-42 polypeptides were fluorescently labeled with Cy2 (Amersham), and the fluorescently labeled polypeptides were added as probes into the cerebrospinal fluid samples of patients to observe the aggregation phenomenon of amyloid β peptides. The experimental results showed that the cerebrospinal fluid samples of 15 Alzheimer's patients showed a "seed aggregation" state, while the 19 age-matched control groups did not show a "seed aggregation" state. The method used in the article can readily detect brain amyloid-beta protein pathology in Alzheimer's disease in vivo and may have potential value for routine diagnosis.

Abstract
Alzheimer's disease is associated with the intraparenchymal growth of plaque-like amyloid deposits 1–3 . Amyloid plaques are formed by the progressive deposition and transformation of soluble amyloid β-protein monomers into insoluble and fibrillar aggregates that contain amyloid β-protein in a β- pleated sheet conformation. This process is described as 'seeded polymerization' of the monomers with slow-nucleation and fast-growth kinetics4. Soluble amyloid β-protein monomers are present in the cortical extracellular space and in the cerebrospinal fluid5,6, whereas insoluble aggregates so far can be found only by the examination of brain tissue by biopsy or autopsy. Here we present a biophysical method that uses the principle of seeded polymerization in combination with fluorescence correlation spectroscopy, which allowed us to detect single amyloid β- Peptide aggregates in the cerebrospinal fluid samples from Alzheimer's patients. All of 15 Alzheimer's samples but none of the 19 age-matched control samples produced large peaks with fluorescence correlation spectroscopy indicating the rapid aggregation of the fluorescent labeled synthetic amyloid β-protein probe onto the amyloid β-protein 'seeds' present in the cerebrospinal fluid. Our method could enable easy  in vivo detection of the cerebral amyloid β-protein pathology of Alzheimer's disease and might be of potential value to facilitate its routine diagnosis.
